    Paediatric drug dosing recommendations from physiologically-based pharmacokinetic (PBPK) modelling

    Generally, drug labels have no recommendations for paediatric use causing clinicians to employ so-called “off-label” practice. Dosing in paediatrics is challenging due to differences in physiology because of organ maturation. Also, ontogeny of drug metabolizing enzymes and transporters may impact drug dosing strategies. A dose cannot simply be based on a child’s body weight and a more sophisticated approach is warranted to ensure safe and effective pharmacotherapy in children. With physiologically-based pharmacokinetic (PBPK) modelling, a virtual child can be build and the plasma concentration of a drug over time can be predicted. With this innovative approach, we may be able to provide paediatric dosing recommendations with enough strength and confidence to eventually discuss our findings with clinicians. In this project, you will determine the feasibility of using PBPK models for establishing dose recommendations and you will investigate whether your model-informed dose for a certain drug and age group can support the dose currently used in paediatric clinical care.

    We’re looking for a student who would like to do his/her internship with us, at the Department of Pharmacology and Toxicology, for at least 12 weeks. All students can reply to this vacancy.
